It depends on the model year and engine type. On 2004 to current model years equipped with the 6.0L diesel, NO engine removal is not necessary to remove the engine oil pan. On 2003 and older equipped with the 7.3L engines, YES it is necessary to remove the engine to access the oil pan. Although the oil pan can be removed if, the transmission is removed, and the engine lifted high enough, resealing it properly would be next to impossible, due to the fact that this particular engine is sealed entirely with silicone from the oil pan to the engine block. The proper way to carry out this repair would be to remove the engine, clean off ALL engine oil from the sealing surface, replace the oil pan sealing it with sufficient amounts of silicone in the front and rear of the engine, where the gap is the widest, and letting it cure overnight before reinstalling the engine and filling up with engine oil.
